چکیده

The replacement of thymine (T) in deoxyribonucleic acid (DNA) by 5-bromouracil (BU) (F. Weygand, A. Wacker, and H. Dellweg, Z. Naturforsch. 7b:19, 1952) is an aid in density separation of types of DNA, and in studies on radiobiology and mutagenesis. The usefulness of BU is, however, limited by its toxic effects. Extensive substitution in DNA of T by BU is lethal for most Escherichia coli and Bacillus subtilis strains. To decrease this toxicity, Oishi and Sueoka (Proc. Natl. Acad. Sci. U.S. 54:483, 1965) suggested adding T to BU-containing media. Because of an apparent heterogeneity

Replication of Bacillus subtilis DNA during germination in 5 bromouracil containing medium and marker mapping.

I N the past few years investigations have been made on DNA replication during germination of Bacillus subtiZis spores in $bromouracil (5-BU) containing medium (YOSHIKAWA 1965; OISHI and SUEOKA 1965; and WAKE 1967). Germination of spores in 5-BU containing medium has also been used for mapping of markers by the synchro-transfer method ( OISHI and SUEOKA 1965 and O’SULLIVAN and SUEOKA 1967). In ...

5-Bromouracil-tolerant mutants of Bacillus subtilis.

5-Bromouracil (BU)-tolerant mutants of Bacillus subtilis 23 (thy his) have been isolated. Several classes of tolerant mutants were obtained by a sequential selection procedure. The classes can be distinguished by their relative BU tolerance as well as several other phenotypic characteristics. The mutants can grow for an extended period of time in minimal medium supplemented with amino acids and...

Tolerance to bromodeoxyuridine in a thymidine-requiring strain of Bacillus subtilis.

Mutations which allow tolerance to 5-bromo-2'-deoxyuridine (BUdR) in a thymidine (TdR)-requiring strain of Bacillus subtilis have been examined. Differences in sensitivity to BUdR existed between isogenic strains harbouring the mutations. Those mutations originally isolated as BUdR-tolerant also bestowed tolerance to 5-bromouracil and vice versa. The strain exhibiting the greatest tolerance to ...

The Bacillus subtilis outB gene is highly homologous to an Escherichia coli ntr-like gene.

The Bacillus subtilis outB gene was found to have strong similarities to an Escherichia coli gene complementing ntr-like mutations in Rhodobacter capsulatus. The deduced gene products had 52% identical amino acids (65% similar residues). The phenotype of strains affected in the OutB function indicates that this B. subtilis gene may be involved in nitrogen utilization.

Bacillus subtilis genes for the utilization of sulfur from aliphatic sulfonates.

A 5 kb region upstream of katA at 82 degrees on the Bacillus subtilis chromosome contains five ORFs organized in an operon-like structure. Based on sequence similarity, three of the ORFs are likely to encode an ABC transport system (ssuBAC) and another to encode a monooxygenase (ssuD). The deduced amino acid sequence of the last ORF (ygaN) shows no similarity to any known protein. B. subtilis c...
